1. IDENTIFICATION
Renal Papillary Antigen-1 (RPA-1) EIA (catalogue # BIO89RPA1).

2. USE
An enzyme immunoassay for the quantitative estimation of RPA -1 in rat urine.
For research use only. Not for use in diagnostic procedures.

3. COMPOSITION
Reagent Content
Coated Strips: 12 x 8 well strips coated with IgG directed
against RPA-1.
Conjugate Concentrate: 1 x 300 uL (50X) anti-RPA-1 HRP-IgG
conjugate. Contains 2% Antibody Stabiliser.
Conjugate Diluent: 1 x 11 mL of conjugate dilution buffer.
Contains Trizma Base, Protein Stabiliser,
ProClin 950, Bronidox L.
Calibrator *: 1 x 500 uL of partially purified rat RPA-1.
Contains ProClin 950, Bronidox L.
Positive Control *: 1 x 100 uL of partially purified rat RPA-1.
Contains ProClin 950, Bronidox L.
Rat Urine Stabilising Buffer: 1 x 10 mL of stabilising buffer. Contains
Protein Stabiliser, Sodium Azide, Thiomersal.
Wash Concentrate: 1 x 55 mL of a concentrated buffer (25X).
Contains 6% Trizma Base, ProClin 950.
Sample Diluent: 1 x 50 mL of ready to use diluent. Contains
Igepal CA-630, ProClin 950.
Substrate: 1 x 11 mL of Tetramethylbenzidine (TMB)
solution.
Stop Solution: 1 x 11 mL of 0.5M Sulphuric Acid (4.9% w/w)
H2SO4.

4. HAZARDS IDENTIFICATION
Sodium Azide is an irritant. Avoid contact with components containing azide. Do
not ingest or inhale. Harmful if swallowed. Contact with acids liberates very toxic
gas.
Sulphuric acid is corrosive. Avoid contact with the skin and eyes. If contact
occurs rinse immediately with water and seek medical advice.
TMB (3,3鈥?,5,5鈥? 鈥? tetramethylbenzidine) may irritate eyes, skin and mucous
membranes. It is a possible mutagen. Any substrate that comes in contact with the
skin should be rinsed off with water.
Trizma Base is an irritant. Irritating to eyes, respiratory system and skin. Remove
contaminated clothing and shoes.
Bronidox L is harmful if swallowed and irritating to skin.
Thiomersal is toxic by inhalation, in contact with skin and if swallowed. It is
irritating to eyes, respiratory system and skin. It is a possible mutagen, which may
affect kidneys and nerves.
ProClin 950 causes burns and sensitisation by skin contact.
Igepal CA-630 is harmful if swallowed and cause serious damage to eyes.

5. FIRST AID PROCEDURES
If exposure is severe remove to fresh air.
Inhalation:
Wash skin with copious amounts of water.
Skin Contact:
Flush eyes with copious amounts of water for at least 15
Eye Contact:
minutes. Assure adequate flushing of the eyes by separating the
eyelids. If any irritation persists, obtain medical assistance.
Wash out mouth with copious amounts of water. Give plenty of

water to drink. Obtain medical attention if large quantity is
ingested.

6. FIRE FIGHTING MEASURES
Wear self-contained breathing apparatus and protective clothing to prevent contact
with skin and eyes.
Some components may decompose and emit toxic fumes under fire conditions
(Sodium Azide, Trizma base, TMB, Thiomersal, Bronidox L, Proclin 950, Igepal
CA-630).

7. ACCIDENTAL RELEASE MEASURES
Environmental precautions: Urine Stabilising Buffer contains Sodium Azide,
which may form potentially explosive metal azides with lead and copper
plumbing. For disposal, the reagent should be flushed with large volumes of water
to prevent Azide build up. The Antibody Stabiliser in the Conjugate Concentrate
is toxic to aquatic organisms. Do not empty in the drains undiluted.
Methods for cleaning up: Wipe up spills with absorbent paper, clean area with a
concentrated chlorine solution and copious amounts of water. Discard all
materials used to wipe up spills using biohazard waste facilities. Avoid contact
with skin and clothing.

8. EXPOSURE CONTROL (PERSONAL PROTECTIVE EQUIPMENT)
The calibrator/positive control and all rat urine specimens are considered
potentially biohazardous materials.
Inhalation: Use with adequate ventilation. Avoid inhalation.
Eye Contact: Wear goggles or safety glasses.
Skin Contact: Wear impermeable gloves and appropriate lab apparel.

9. HANDLING AND STORAGE
Handling: All rat urine specimens and calibrator/positive control material should
be handled as though potentially biohazardous. With components do not breath in
vapours and do not get in eyes, on skin, on clothing.
Specific Use: For research use only. Not for use in diagnostic procedures.
Storage: Store all reagents and ELISA plates at 2-8oC.
Positive Control and Calibrator store at -20 oC


10. PHYSICAL AND CHEMICAL PROPERTIES

11. STABILITY AND REACTIVITY
Stop Solution (0.5M H2SO4) is incompatible with water and bases.
Urine Stabilising Buffer contains Sodium Azide, which may form potentially
explosive metal Azide with lead and copper plumbing. Sodium Azide explodes
when heated and may form hazardous combustion or decomposition products.
Substrate (TMB solution) is incompatible with strong oxidising agents.
Thiomersal is incompatible with strong oxidising agents, strong acids and strong
bases.
Bronidox L is incompatible with strong oxidising agents.
Proclin 950 is incompatible with strong oxidising agents, Amines, Mercaptans and
Reducing agents.

12. TOXICOLOGICAL INFORMATION
One reagent contains Sodium Azide, which is toxic by inhalation, in contact with
skin and/or if swallowed. Proclin 950 may cause allergic skin reaction and is
extremely destructive to tissue of the mucous membranes and upper respiratory
tract, eyes, and skin.

13. ECOLOGICAL INFORMATION
No known ecological hazards.

14. DISPOSAL GUIDELINES
Observe all Federal, State and Local laws concerning health, disposal and
pollution. Dispose of all specimens and components according to established
safety procedures.

15. TRANSPORT INFORMATION
This manufactured product is not subject to the International Air Transport
Association Dangerous Goods Regulations.
